Optimization of conditions for increasing of saffron cell biomass and crocin production in stirred bioreactor

Bioreactors provide suitable conditions for the growth of cells and production secondary metabolites by regulating physical chemical factors. In this study, saffron cell crocin metabolite were evaluated in Erlenmeyer flask a stirred bioreactor. To optimize conditions, effect different concentrations 2-(N-morpholino) ethanesulfonic acid (MES) (0, 2.5 5 mM) as buffering agent sucrose (3%, 6% gradually 6%) addressed on flask. This aim was then pursued bioreactor through investigation aeration pH medium adjustment. The results first step experiment showed that Schenk Hildebrandt basal with naphthalene acetic (2 mg l−1) 6-benzylaminopurine (1 supplemented mM MES well gradual increment from 3 to caused highest biomass production. spectrophotometry measurement content 0.8 g−1 after 6 weeks. second part revealed bioreactor, constant (5.8) during period is limiting factor Aeration also found be an inhibiting crocin. that, if evaporated volume water constituted, it can effective increase around 2 folds. addition, total proliferated system under non-constant without could raised up dry weight, based HPLC determination.
